Molecular identification of isolated microorganisms of artisanal cheese making shop located in la Libertad-Chontales, Nicaragua

In the artisanal cheese production of Nicaragua, a number of microorganisms coexist that have not been studied, and many times it is not considered how they intervene in the production of cheese. For this reason, in this work microorganisms were isolated from an artisan cheese making shop in basic culture media AN, LB, PCA, PDA and AM in order to isolate a large number of microorganisms. Once the cultures were isolated and purified, 82 bacteria were obtained, 3 yeast fungi and 12 filamentous fungi. By means of the morphological analysis of the macro and microscopic characteristics, 16 morphotypes that included bacteria and fungi were selected for molecular identification by means of DNA extraction, PCR amplification and sequencing of the 16S regions for bacteria, ITS for filamentous fungi and the D1 / D2 / D3 domains for yeast. 20 isolates were identified at the species level: Bacillus cereus (1), Enterobacter cloacae (1), Escherichia coli (1), Klebsiella pneumoniae (2), Planococcus maritimus (1), Proteus vulgaris (1), Pseudomonas aeruginosa (2 )), Psychrobacter Alimentarius (1), Serratia marcescens (3), Staphylococcus aureus (1), Cybelindnera jadinii (2), Rhodotorula mucilaginosa (1), Candida paraugosa (1), Aspergillus hiratsukae (1) and Trichoderma orientale (1) . In addition, 7 bacteria are identified at the genus level: Enterobacter sp (4), Escherichia sp (1), Klebsiella pneumoniae (1) and Pseudomonas sp (1). This research contributes to the advancement of the knowledge of the microorganisms present in the production process of an artisan cheese maker, allowing the selection and use of these for their application in biotechnological processes.
